Abstract Seed size had an important effect on rate and final germination in Zea mays and Soya bean. Also seed size effected growth in these plants. Zea mays and Soya bean plants were selected for this study for their economical importance as source of food for human and animals. This study was carried out in laboratory as well in the field. The effect of different temperatures was deter mined on rate and final germination in both seeds used. Also effect of light on seed germination was tested. The results revealed that there is big effect of seed size on germination and subsequent growth of plants used. Where larger seeds gave better rate and final germination, there was better growth in plants formed from larger seeds. The best temperature for germination is 25ْc for the two types of plants used. Light had little promoting effect on rate and final germination of seeds used .Seed germination determinted in the soil under field conditions. The results obtained agreed with laboratory results but in Soya bean, it was found better germination from small seeds. This is may be due to seed coat being weaker and thinner in small seeds than big seeds. Different categories of growth was measured as plant length, fresh weight leaf weight dry weight etc. The results were statistically analysed and discussed.Different Growth categories were determined. These were better in plants originated from large seeds than small seeds. We can classify our results as increasing the size and weight of seed gave better results in germination, growth and length of plants, fresh weight and dry weight.

Sclerotinia sclerotiorum "

Abstract This study was conducted to evaluate in vitro and in vivo antagonistic activities of five Trichoderma isolates; three local (Lib1, Lib2, Lib3) and two commercial (T22, Tp1) against mycelium radial growth and sclerotia formation of the plant pathogenic fungus Sclerotinia sclerotiorum, and its effect on seed germination and infection of pea (Pisum sativum L.) seeds. All experiments were conducted at two growth temperatures, 20 and 25˚C. Microscopic interactions of Trichoderma isolates and host fungus were performed by slide culture technique. Two concentrations (10, 20%) of Trichoderma culture filtrate were tested for their effect on mycelium growth. All Trichoderma isolates were able to inhibit the growth of the fungal pathogen, although, variations were observed among tested isolates in rate and mechanisms of inhibition. At 25⁰C Lib2 showed the highest rate of mycelium growth and spore formation. In mycoparasitism, the inhibition rate was 55% for Lib3 and 45% for Lib1. Whereas for indirect interactions; significant inhibition of mycelium radial growth was observed for Lib1 and Tp1 with a percentage of 28.5 and 26.4 respectively. Overgrowth was also reported on the colonies of S. sclerotiorum by Lib2 and T22, and these two isolates showed remarkable inhibition of sclerotia formation with a percentage rate of 91. The effect of culture filtrate of Trichoderma isolates was dose dependent, 20% concentration of culture filtrate lowered the rate of radial growth of S. sclerotiorum by 53% for Lib1 and 47.2% for Tp1, while lower inhibition of Sclerotinia radial growth was reported for 10% concentration of culture filtrate with a percentage of 45 for Lib1 and 34 for Tp1. At 20⁰C 10% and 20% culture filtrate exhibited lower effect on mycelium radial growth of Sclerotinia. Seed germination of peas (local variety) was enhanced by Trichoderma treatment. Increased rate of seed germination was reported by 96.6% for Lib1, Lib2 and 80% for Lib3 and 76% for Tp1, lower inhibitory effect was reported for T22 by 43%. Seed treated with Trichoderma isolates exhibited low rate of seedlings infection by S. sclerotiorum.

Abstract This study was carried out during 2009-2010 in plant tissue culture laboratories of Biotechnology Research Centre and Laboratories in Al-Fatah University in departments of Botany and Horticulture with cooperation of the the Agricultural Research Centre. The aim of the study was the conservation through micropropagation of the plant species Balanites aegyptiaca (L) Del which is facing extinction from the Libya desert habitat. Samples including branches and seeds of wild Balanites aegyptiaca were collected from plants growing desert valleys around the oasis of Ghat during winter, 2009. Seed dormancy and viability were tested as well as some of the micropropagation methods including indirect micropropagation by formation of callus cells and direct micropropagation under the effect of several concentrations of a number of plant growth regulators. The results showed that the seed viability was between 88 and 90%, moreover, the seeds were under two dormancy stages, internal and external. The best treatment to overcome the external dormancy was through getting the seeds out of the endocarp by a mechanical treatment. The internal dormancy, however; was overcome by cultivating the seeds on MS medium supported by Gibberellic Acid at a concentration of (2 mg/L); this treatment revealed a germination percent of 90%.This study also included the effect of the type of explant on response percentage and weight of the callus cells formed. The highest percentage of callus cells formed was obtained from leaf explants cultured on MS medium containing 2, 4-D in a concentration of (2 mg/L), where the response was 100%. Cotyledon explants recorded highest callus cells of mean weight of (1.979 g) per callus. This study also has illustrated that some of the root tips incubated in dark at MS medium free of plant regulators have directly differentiated into shoots. The results of this study have demonstrated that the best effect of Benzyl adenine (BA) plant regulator on direct differentiation of root explants was at 0.5mg/L for all the three studied parameter; which were the mean of shoot number (1.66), mean of shoot length (1.38 cm) and mean of number of formed leaves (1.47). The results of the culture of single node on MS medium containing BA (0.5mg/L) have shown that the highest mean of shoot number and shoot length were 2.9 shoots and 5.15 cm in length. Whereas, the highest mean of number of leaves (15.75 leaves) was recorded when 1mg/L of BA was used. The effect of Kinetin (Kin) on differentiation of single nodes revealed that the concentration of 1.5 mg/L has produced the highest mean of shoot number (2.5 shoots), shoot length (3.25 cm) and leave number (14.7 leaves), Results have also revealed that the rooting percentage wqs 100% when using ½ MS (Murashig & Skoog). Survival percentage of plantlets has reached 80% under a plstic green house with no control of the environmental conditions.A summary of the best micropropagation protocol of treatments from fruit (seeds) to acclimated plantlets is illustrated in Figure (31).
